Depends on what board you are trying the hotflash for. What is the make/model?

You need a mainboard with socketed bios, replacement bios chips, tweezers, a small screwdriver with flat blade and a bootmedium with an OS, a flasher and the bios file obviously.

once the mainboard is showing the bios screen, you can remove the original bios chip, since it has been copied to RAM already. or just wait until your bootmedium has booted to a prompt.
be careful not to short anything while removing the bios chip. use the flat blade scewdriver for DIL and the tweezers for PLCC chips. carefully insert the replacement chip and start the flash tool. make sure the flashtool recognizes the chip correctly and flash it. after flashing you can safely switch off the PSU, pull out the chip and replace it with the original. Done :)

And I can do this no matter what board I have?

make sure the flashtool recognizes the chip correctly and flash it.
this means you need to check if the bios chip will fit in the socket of the "flashing" board and if the voltage/size is support by the "flashing" board. Usually if you have boards from the same period/with same line of processors this will be the case.
